I set up a direct debit in September. It is showing as active but no payment was taken this month (October), does it take a while to set up?

There can be a small delay when setting up a new Direct Debit. When you set up a Direct Debit for the first time it won't collect until the following months payment, meaning the current outstanding payment would need to be covered manually.
